    Observing RNA polymerase at work in real time

    RNA Polymerase II is known to be largely responsible for the precise control over gene expression for each specific cell lineage. Until recently, a multidisciplinary team from Stanford discovered its detailed mechanism in gene expression as they observed this enzyme transcribing DNA in real time. A handmade preinitiation complexs (PIC) was assembled by expressing 32 different proteins from yeast, with the resulted machinery fully functional at the single-molecule level. As this PIC initiated polymerase II activity and began to transcribe a template DNA, the pulling motion affected the laser light scattering on the DNA strands and allowed measurement down to the nanometer level.
